HLA tetramers, also known as MHC tetramers, are the gold standard for precise detection of CD8+ antigen-specific T cells. Widely used in the development of anti-cancer and antiviral immunotherapies, they enable detailed analysis of immune responses.

 

 

HLA refers to human leukocyte antigen, part of the MHC gene complex in humans, which presents peptides from pathogens and self-antigens on cell surfaces. These peptides, in combination with HLA, are surveilled by T cells via their T cell receptors. If recognized as foreign, these cells are targeted for destruction by the immune system.

 

Recombinant HLA tetramers, linked to peptide antigens and fluorochrome-conjugated streptavidin, are used to detect antigen-specific T cells via flow cytometry.
